Link building is the act of acquiring links from other websites, with the end goal of boosting the search engine ranking of your own website. "Link popularity" refers to the number and quality of inbound links from other websites and blogs to your own site. This is one of things search engines use to evaluate websites (and rank them), so it's an important topic for most webmasters.
